陈皮橙皮苷提取 高效液相色谱法测定陈皮健胃片中橙皮苷的含量

[摘要]意图 树立陈皮健胃片中橙皮苷含量的测定办法。办法 选用高效液相色谱法（HPLC），色谱条件为Inertsil ODS-SP（4.6 mm×150 mm，5 μm）色谱柱别离，检测波长为283 nm，活动相为乙腈-水进行梯度洗脱，流速为1.0 ml/min，柱温为30℃，理论板数≥3000。成果 橙皮苷进样量线性规模为0.10～2.00 μg；在精密度试验、稳定性试验、重复性试验中，橙皮苷峰面积的相对标准偏差（RSD）为2.06%、2.12%、2.14%；橙皮苷的加样回收率为97.36%～102.39%，平均值为99.29 %，RSD为2.06%（n=9）。定论 本法的重现性和专特点杰出，加标回收率符合要求，能精确、稳定地对陈皮健胃片中的橙皮苷含量进行定量测定，可作为该制剂的质量操控办法之一。

[Abstract]Objective To establish a method for the determination of hesperidin in Chenpi Jianwei tablet.Methods A high performance liquid chromatography （HPLC） method was established.The chromatographic conditions were as follows：the chromatographic column Inertsil ODS-SP （4.6 mm×150 mm，5 μm） with the mixture of acetonitrile and water as mobile phase in gradient mode，the detection wavelength at 283 nm，the flow velocity of 1.0 ml/min，the column temperature at 30 ℃，the theoretical plate number no less than 3000.Results The linear relationship between content and peak area was obtained during the range of 0.10–2.00 μg for hesperidin.RSD of the peak area of hesperidin for the precision test，the stability test and the repeatability test was 2.06 %，2.12 % and2.14 % respectively.The recovery rate of hesperidin obtained were in the range of 97.36%–102.39% and the average of 99.29% with RSD of 2.06% （n=9）.Conclusion This method can be applied to quantitative assay of hesperidin in Chenpi Jianwei tablet with good specificity and reproducibility.The sampling recovery test is in compliance with the requirement，which can be applied to the quality control of Chenpi Jianwei tablet.

陈皮健胃片处方源于第四批全国老中医药专家学术承继作业指导老师，硕士生导师余伯亮主任中医师经验方，由广陈皮、山楂、淮山等组成。该方理气和胃，健脾消食，用于脾胃衰弱、消化不良等症。为便于患者服用，丰厚临床用药剂型，研制成陈皮健胃片。研讨发现，柑橘属植物中含有丰厚的黄酮类化合物，有文献[1]报导，橙皮苷是该植物中重要的活性成分，具有铲除氧自由基[2]、抗肿瘤[3]、抗病毒[4]、抗炎[5]、下降血脂、维护心血管[6]等多种成效。

橙皮苷含量测定办法有分光光度法[7-8]、荧光光谱法[9]、高效毛细管电泳法[10-11]、高效液相色谱法[12-14]、超高效液相色谱法[15-16]、薄层扫描色谱法[17-18]，为有用操控陈皮健胃片的产品质量，确保安全有用地用药，本研讨经过HPLC测定陈皮健胃片中的橙皮苷含量，并对3批克己陈皮健胃片中的橙皮苷进行含量测定。

1资料与仪器

1.1资料

陈皮健胃片（批号：20150401、20150410、20150420）由本试验室克己；橙皮苷（我国药品生物制品检定所）：批号110721-200613；色谱醇甲醇和乙腈。试验用水为双蒸水，所用其他试剂均为剖析纯。

1.2仪器

Ultimate 3000高效液相色谱仪（美国戴安公司）；CPA225D电子天平（德国赛多利斯公司）；KQ-300VDE超声波清洗仪（昆山市超声仪器有限公司）；U-3010分光光度计（Hitachi Corp.）。

2办法与成果

2.1含量测定目标的断定

陈皮为方中君药，在粤被称为广陈皮或新会陈皮。橙皮苷、黄酮类化合物等为其主要成分。其对某些革兰氏阳性菌、真菌有较好的抑制效果[19]，对小鼠胃排空有促进效果，不会明显影响阿托品所引起的胃排空抑制效果和胃复安所发生的胃排空强化效果[20]；其水煎剂对家兔离体十二指肠梗阻自发活动有较好的抑制效果，可以下降其缩短力，缓解其严重性，并呈现量效之反响联系[21]。该君药经过对胃肠道平滑肌功用的改动，使胃肠道上端正常运动得以康复，然后起到健胃效果，故本研讨以橙皮苷作为陈皮健胃片质量操控的目标成分，树立了HPLC测定陈皮健胃片中橙皮苷含量的办法。
